بررسی تغییرات بیان ژن‌های HSP60 و HSP70 متعاقب تمرین بی هوازی با شدت بالا پس از ایسکمی – رپرفیوژن در عضله قلبی رت های صحرایی نر: اثر مکمل ملاتونین

چکیده

زمینه و هدف: هدف از پژوهش حاضر بررسی تغییرات بیان ژن HSP60 و HSP70 متعاقب تمرین بی هوازی با شدت بالا در عضله قلبی رت های صحرایی نر پس از ایسکمی – رپرفیوژن: اثر مکمل ملاتونین بود.
مواد و روش ها: : بدین منظور 24 رت نر صحرایی ویستار با وزن تقریبی 250-200 گرم به 4 گروه 6 تایی شامل گروه کنترل، ملاتونین، تمرین بی‌هوازی، و تمرین بی‌هوازی + ملاتونین تقسیم شدند. پروتکل تمرین بی‌هوازی به مدت یک ماه اجرا شد؛ ضمنا گروه‌ تمرین بی‌هوازی ملاتونین در مدت تمرین نیز تحت عمل گاواژ ملاتونین قرار داشتند. در پایان یک ماه رت‌ها بعد از دو روز استراحت تحت تزریق داروی ایزوپرنالین برای ایجاد ایسکمی ریپرفیوژن قرار گرفتند. بعد از دو روز رت‌ها تشریح و بطن چپ آنها خارج گردید. سپس میزان بیان ژن HSP60 و HSP70 رت‌ها با استخراج RNA و با تکنیک Real Time اندازه‌گیری شد.
یافته ها: نتایج تحقیق نشان داد که مصرف ملاتونین میزان بیان ژن آپوپتوزی HSP60 زا کاهش و HSP70 را پس از تمرین بی هوازی افزایش داد (05/0≥p).
نتیجه گیری : نتایج تحقیق حاضر نشان داد که تمرینات بی‌هوازی شدید با مصرف ملاتونین موجب کاهش حجم آپوپتوز میوکارد پس از ایسکمی ریپرفیوژن ناشی از تزریق ایزوپرنالین می‌شود.
