بررسی اثرات سیتوتوکسیک در متابولیت‌های تولیدشده به‌وسیلۀ اکتینومیست‌ها

چکیده

زمینه و هدف: با توجه به افزایش شیوع و گستردگی سرطان، امروزه نیاز به داروهای شیمی­درمانی سرطان هرچه بیشتر احساس می­شود. محصولات طبیعی باکتریایی منبعی بسیار غنی برای ترکیبات جدید با فعالیت­های زیستی مختلف هستند. در میان باکتری­ها، اکتینومیست­ها پتانسیل قابل توجهی را در تولید داروهای ضد سرطان دارند. در این پژوهش، اکتینومایست­های مولد ترکیبات سیتوتوکسیک غربالگری شده‌اند.
روش بررسی: 40 سویه اکتینومایست مختلف جداشده از مناطق مختلف کشور از مرکز کلکسیون میکروارگانیسم­های دانشگاه تهران دریافت شد و پس از احیاء، مراحل پیش تخمیر و تخمیر و استخراج متابولیت­های ترشحی آنها انجام شد. میزان سمیت عصاره­ها بر آرتمیا، به­عنوان آزمون سریع و ساده برای غربالگری ترکیبات ضد سرطان سنجیده و عصاره ها در سه گروه بسیار سمی (40-59 درصد)، نیمه سمی (20-39 درصد)، غیر سمی (0-19 درصد) دسته­بندی شد.
یافته‌ها: از بین40 سویۀ مطالعه­­شده، که تولید آنها مورد بررسی قرار گرفت، 23 سویه بسیار سمی ،16 سویه نیمه سمی و 1سویه با سمیت کم بوده‌اند.
نتیجه­گیری: در این پژوهش نشان داده شد که اکتینومیست­ها‌ی جداشده از مناطق مختلف ایران می­توانند منبعی مناسب برای تولید ترکیبات سیتوتوکسیک باشند. این ترکیبات می­توانند به­عنوان ترکیبات ضد سرطان برای کارهای بعدی در نظر گرفته شوند.
