بررسی تاثیر دوز بالای اسید آسکوربیک بر فعالسازی مسیر نکروپتوز در رده سلولی HT29 سرطان کولون

چکیده
هدف: مطالعه به بررسی تاثیر دوز بالای اسید آسکوربیک بر بیان ژن های کلیدی مسیر نکروپتوز در سلول های HT-29 سرطان کولون اختصاص یافت. مواد و روش ها: سلول های HT-29 در محیط کشت DMEM-HG حاوی FBS 10% و Pen/Strep 1% کشت داده شدند و در شرایط دمای 37 درجه سانتی گراد حاوی CO2 5% انکوبه شدند. سلول ها پس از رسیدن به تراکم 90% تریپسینه شدند و با تراکم 30000 سلول در هر چاهک بصورت سه بار تکرار در پلیت های کشت دوازده خانه کشت داده شدند. سلول ها به دو گروه تقسیم شدند. گروه کنترل که تنها محیط کشت پایه دریافت میکرد و گروه تست که به مدت 3 ساعت تحت تیمار با اسید آسکوربیک 5 میلی مولار قرار می گرفت. پس از 24 ساعت محیط رویی سلول ها و رسوب سلولی جهت بررسی های بعدی جمع آوری شد. درصد آپوپتوز و نکروز به روش فلوسیتومتری و اندازه گیری فعالیت LDH به روش کینتیک انجام شد. آنالیز بیان ژن ها به روش Real time PCR صورت گرفت. آنالیز آماری با استفاده از ورژن 8 نرم افزارgraph pad prism و t-test انجام شد. نتایج: تصاویر میکروسکوپی و نتایج فلوسیتومتری موید القای نکروپتوز توسط اسید آسکوربیک بودند. بعلاوه، تیمار با اسید آسکوربیک میزان بیان ژن های کلیدی نکروپتوز و فعالیت LDH را بطور معنی دار افزایش داده بود. نتیجه گیری: دوز بالای اسید آسکوربیک می تواند بعنوان عامل فعال ساز نکروپتوز در سلول های HT- 29 مطرح باشد.
