The effect high and low intensity training on muscle density, SERCA2a and phospholamban in slow-twitch and fast twitch muscles of male Wistar Rat

Abstract
Background and Objective. The present study aimed to investigate the inaction effect of exercise intensity and fiber type on the SERCA, major transmitter of calcium, and its inhibitor’s factor, PLN.
Methods. 36 adult male Wistar rats (325 ± 35 g) were randomly divided into two low intensity (LIT=12) and high intensity training (HIT=12) groups and one control group (CO=12). The exercise was performed at a speed of 12 to 15 (m / min), and 25 to 30 (m / min), respectively, in the low intensity and high intensity group, between each period, the active rest was at a speed of 10 (m / min), Exercises were performed 5 days a week for 8 weeks. At the end, the EDL and Sol muscles were isolated, centrifuged, and converted to serum. The ELISA kit and the biotech analyzer ELISA were used for analysis. ANOVA, t student and factor analysis were used for analysis of the data.
Results. The main effect of exercise intensity in SERCA2a and PLN proteins density was significant only in the EDL muscles. And also interaction effect between exercise intensity and fiber type was significant for SERCA2a and PLN proteins density (p=0.05). Generally, Ratio of inhibitory factors of PLN to SERCA2a densities was decreased in the LIT and HIT groups than control group by 29% and 4%, respectively.
Conclusion. HIT method have a more effect on increasing of SERCA2a function in the EDL muscles
