CA can inhibit the growth of various human tumor cell lines such as colorectal, leukemia, hematoma, as well as melanoma. This study aims to investigate the effect and action mechanism of CA on apoptosis of MCF-7 breast cancer cells. Subjects and Methods:MCF-7 breast cancer cells treated with different concentrations of CA (0, 10, 50, 100 and 200 mM) for 24 h. Cell viability was determined using MTT assay. Apoptosis markers including DNA fragmentation, phosphatidylserine exposure at the outer leaflet of the plasma membrane measured using TUNEL and flow cytometery and Annexin V affinity. Results: Viability assay showed that the IC50 of CA in MCF-7 cells was about 50 mM. CA at dose of 100 mM significantly induced the apoptosis after 24 h treatment as assessed by TUNEL+ cells and evidence of late apoptotic cells with Annexin V+ and propium iodide (PI+). CA can inhibit the growth of MCF-7 cells by inducing cell apoptosis. The results can be helpful in understanding the anticancer mechanism of CA and gain further support for its safe use as an alternative and complementary drug in cancer therapy.}, keywords = {Apoptosis,MCF-7 Cancer cell line,Trans cinnamic acid}, title_fa = {تأثیر ترانس‌سینامیک اسید بر شاخص‌های آپوپتوزی در سلول‌های سرطانی رده ی MCF-7}, abstract_fa = {زمینه و هدف: ترانس سینامیک اسید (CA)، محصول دآمیناسیون فنیل­آلانین در تمام بافت­های گیاهی و جزء اصلی و فعال مواد غذایی گیاهی مورد استفاده انسان است. ترانس­سینامیک می­تواند رشد سلول­های تومور های مختلف انسان مانند سرطان روده بزرگ، لوسمی، تومور کبدی و همچنین ملانوم را مهار کند. هدف از این مطالعه بررسی اثر و مکانیسم عمل ترانس سینامیک بر آپوپتوز سلول MCF-7 سرطان پستان است. روش بررسی: به منظور بررسی اثر و مکانیسم عمل ترانس سینامیک، سلول های سرطان سینه با غلظت های مختلف ترانس‌سینامیک (10،0، 50، 100 و 200 میلی­مولار) به مدت 24 ساعت تیمار شدند. زنده­مانی سلول­ها با استفاده از روش MTT بررسی شد. شاخص­های آپوپتوز شامل قطعه قطعه شدن DNA و تظاهر فسفاتیدیل­سرین در لایه بیرونی غشای پلاسمایی به ترتیب با استفاده از تست­های TUNEL و فلوسایتومتری و ارزیابی اتصال Annexin V، ارزیابی گردید. یافته­ها: مقدار LC50 ترانس‌سینامیک در سلول­های MCF-7 بر اساس ارزیابی زنده­مانی به روش MTT، حدود 50 میلی­مولار تعیین شد. تیمار سلول­ها با دوز 100میلی­مولار ترانس‌سینامیک سبب القا معنی­دار شاخص­های آپوپتوز گردید. بطوری که سلول­های TUNEL+ و سلول­های مراحل انتهایی آپوپتوز Annexin V+ و Propium Iodide (PI+) بطور معنی­داری افزایش یافتند. نتیجه­گیری: نتایج این تحقیق نشان داد که ترانس سینامیک اسید با القای آپوپتوز دارای فعالیت ضد­توموری موثری در برابر سلول­های MCF-7 است. این نتایج می­تواند در درک بهتر مکانیسم ضد­سرطانی ترانس سینامیک و تایید بی­خطر بودن استفاده آن به عنوان یک داروی جایگزین و مکمل در درمان سرطان مفید باشد.}, keywords_fa = {آپوپتوز,ترانس‌سینامیک اسید,سلول‌های سرطان سینه MCF-7}, url = {https://jsmj.ajums.ac.ir/article_45396.html}, eprint = {https://jsmj.ajums.ac.ir/article_45396_7705e5c307600a66f9eda4e9af661ccc.pdf} }
