بیان نوترکیب انتروتوکسین استافیلوکوکوس اورئوس تیپ B به عنوان کاندیدای واکسن

چکیده

زمینه و هدف: استافیلوکوکوس اورئوس(Staphylococcus aureus)، کوکسی گرم مثبتی است که می­تواند بیماری های مختلفی چون مسمومیت­های غذایی، شوک های مخاطره آمیز و ... را به وجود آورد. از مهم­ترین سموم تولید شده توسط این باکتری، انتروتوکسین نوع B یکی از معمولترین و مهم­ترین توکسین­های یافت شده و یک سوپر آنتی ژن می باشد. هدف از انجام این تحقیق همسان سازی و بیان این پروتئین به شکل نوترکیب جهت دستیابی به یک ساختار و منبع پایدار برای تولید آن به منظور بررسی های آینده به عنوان کاندید واکسن بود.
روش بررسی: ژن seb به لحاظ وجود کدون­های نادر مورد بررسی قرار گرفت و بهینه سازی ژن با استفاده از نرم افزارهای بیوانفورماتیکی انجام شد. واکنش هضم آنزیمی برای تایید حضور ژن نوترکیب  seb درون وکتور بیانی(+)pET28a انجام شد. پس از تأیید همسان سازی ژن مورد نظر، بیان نوترکیب پروتئین SEB با استفاده از القاءگر مصنوعی IPTG و همچنین تخلیص تحت شرایط طبیعی(غیردناتوره) با کمک ستون کروماتوگرافی نیکل(Ni-NTA) صورت پذیرفت. آنالیز وسترن بلات نیز برای تأیید پروتئین SEB انجام شد.
یافته­ ها: شاخص سازگاری کدون(CAI) مربوط به ژن طبیعی 68/0 بود، درحالی که این شاخص برای ژن بهینه سازی شده به 82/0رسید. درصد کدون­های با شیوع بالا در توالی بهینه شده به 57 درصد افزایش یافت. هضم آنزیمی صحت همسانه سازی ژن seb در وکتور (+)pET28a را تائید کرد. نتایج نشان داد که همسانه سازی ژن seb درون وکتور (+)pET28a در یک جایگاه مناسب بین محل­های مورد انتظار اثر آنزیم­های برشی انجام شده بود. همچنین ژن seb بعد از القاء با IPTG بیان خوب و قابل توجهی داشت. میزان پروتئین بیان شده برای هر لیتر از محیط کشت حدود 22 میلی­گرم بود. وسترن بلاتینگ، واکنش پروتئین نوترکیب با آنتی بادی ضد هیستیدین را نشان داد.
نتیجه­ گیری: سازه نوترکیب پایدار محتوی ژن seb تولید گردید که بیان نوترکیب قابل توجهی از پروتئین را نشان داد. بنابراین پروتئین SEB را می توان در مطالعات آینده برای بررسی میزان ایمن سازی علیه سم SEB مورد بررسی قرار داد.
