اثر عصاره گیاه Euphorbia microsciadia Boiss بر تکثیر سلولی و بیان سایتوکاین ها در سلولهای سرطان پستان رده MDA-MB-231

چکیده

زمینه و هدف : E. microsciadia در بسیاری از کشورها در طب سنتی برای معالجه سرطان استفاده می شود. مطالعه حاضر با هدف تعیین اثر عصاره E. microsciadia بر کشندگی سلول های توموری و بیان سایتوکاین ها در رده سلولی MDA-MB-231 طراحی و اجرا شده است.
روش بررسی : سلول ها در محیط DMEM به همراه ۱۰ درصد سرم جنین گاوی، حاوی پنی سیلین و استرپتومایسین کشت داده شدند. اثر عصاره بر زیست پذیری سلولهای MDA-MB-231 با روش MTT بررسی شد. برای ارزیابی میزان بیان mRNA سیتوکاین ها بعد از تیمار رده سلولی با عصاره RNA استخراج شد. بعد از استخراج RNA و تبدیل آن به cDNA، میزان بیان mRNA در نمونه ها توسط تکنیک TaqMan real-time PCR اندازه گیری شد.
یافته ها: پس از 24 و 48 ساعت مقادیر IC50 برای یوفوربیا میکروسیدا 30/278 و 20/238 میکروگرم در میلی لیتر بود. عصاره اثرات ضد تکثیری در رده سلولی MDA-MB-231 به روش وابسته به دوز و زمان به نمایش گذاشت. INF-γ IL-8, IL-1β, افزایش بیان معناداری 24 و 48 ساعت بعد از تیمار با عصاره نشان دادند. TGF-β1, IL-6, IL-10, IL-17A بعد از 24 ساعت افزایش بیان و پس از 48 ساعت از تیمار کاهش بیان نشان دادند.
نتیجه گیری: عصاره ی E. microsciadia ممکن است بتواند به عنوان ابزاری برای دست کاری بیان ژن های سایتوکاین ها پیش گفته و در نهایت، کنترل رشد و تکثیر سلول ها که امری اساسی در مطالعات ملکولی و سلولی حیطه ی سرطان پستان است شود.
